Atsuete Solution for Biological Staining (Abstract)

Annatto


This study was conducted to determine and analyze the ability of Bixa Orellana (atsuete) solution in determining which atsuete solution (75 %, 50%, and 25%) can distinguish the parts of a human cheek cell and onion cell clearly.


The making of annatto solution is done manually by putting the different number of annatto whether it is 75%, 50% and 25% on the same kind of container. Then different amount of water (75%, 50% and 25%) was put on each container. Then the atsuete was soaked in the water for some minutes until all the color of it was already on the water. It was dropped on the thin piece of an onion and human cheek cell. It was viewed in the microscope and the data were gathered.


Results under the microscopy method showed that if 75% of the atsuete and 25% water were used to see the specimen, it is better to use because the parts of the cell has a clearer view in this concentration. While in using the 50 % atsuete and 50% water, it is not that clear but you can still view the cells of it. And if you use the 25% atsuete and 75% water, you cannot determine well the parts of the cell because it is light in color and it is not quite visible.


It was concluded that using 75% atsuete and 25% water is the best way in viewing the parts of a human cheek cell and onion cell.
